米カリフォルニア州サンタモニカ出身。父は演出家のレオ・ペン、母は女優のアイリーン・ライアン、兄マイケルは音楽家、弟クリスも俳優という芸能一家で育つ。1981年「タップス」で映画デビューし、「初体験 リッジモント・ハイ」（82）や「俺たちは天使じゃない」（89）に主演。91年には「インディアン・ランナー」で監督・脚本家デビューし、「クロッシング・ガード」（95）などでメガホンをとる。数多くの名匠監督に愛され、「デッドマン・ウォーキング」（95）でベルリン国際映画祭の銀熊賞、「シーズ・ソー・ラヴリー」（97）でカンヌ国際映画祭の男優賞、「キャスティング・ディレクター」（98）と「21グラム」（03）でベネチア国際映画祭のボルピ杯を受賞。「ギター弾きの恋」（99）、「I am Sam アイ・アム・サム」（01）でアカデミー主演男優賞にノミネートされ、「ミスティック・リバー」（03）と「ミルク」（08）で同賞を受賞。現代アメリカ映画を代表する俳優となった。以降の出演作に「きっと ここが帰る場所」（11）、「LIFE!」「L.A. ギャング ストーリー」（ともに13）、「博士と狂人」（19）など。
